- 2 x 250 ml of Agar base
- 400 ml of buffered liquid medium:
This component is provided in excess to allow for individual batches. - 2 x 50 ml heat-inactivated fetal calf serum (iFCS)
- 2 ml of Hemin stock solution, 500x (M-ML-108S):
0,25% solution of Hemin in 30% Triethanolamine, sterile - 5 ml of Pen-Strep stock solution, 200x (M-ML-105S):
10.000 units of penicillin (base) and 10.000 μg of streptomycin (base)/ml as penicillin G sodium salt and streptomycin sulfate in 0.85% saline, sterile - 40 pcs. Nitrocellulose membranes diameter 82 mm:
for easy plating, fluorescence monitoring and transfer of colonies to alternative conditions (e.g. induction of the LEXSY T7 polymerase-based gene expression) - 40 pcs. Petri dishes diameter 90 mm
- 40 pcs. Plating spatula in single bags

Preparation of 500 ml LEXSY plating medium (20 plates, each 25 ml) :
- Microwave-melt 250 ml Agar base in provided glass flask and keep at 55 °C.
- Add pre-warmed (37 °C) buffered liquid medium to ca. 400 ml.
- Add 50 ml iFCS, 1 ml Hemin stock and 2.5 ml Pen-Strep.
- Fill with buffered liquid medium to 500 ml.
- Add appropriate antibiotics (not provided).
- Pour plates with 25 ml pre-warmed (55 °C) serological pipette, avoid air bubbles.
- After solidification, dry the plates open under the laminar flow box for exactly 10 min, than store them closed bottom-up.
- After plating, seal plates with parafilm and incubate bottom- up for 5 - 9 days at 26 °C until colonies appear.
Prepare the plates freshly on the day of plating!
